IDENTIFICACIÓN DE MELOIDOGYNE JAVANICA NEMATODO AGALLADOR EN BEGONIA CULTIVAR COCKTAIL
Esta investigación presenta la identificación de la especie del nemátodo agallador Meloidogyne procedente de raíces agalladas de begonia cultivar Cocktail que crecieron en condiciones de invernadero. La tinción de las raíces con agallas se utilizó el método de Goodey, después bajo el microscopio estereoscópico las hembras de Meloidogyne fueron removidas de un fragmento de raíz. Las hembras de Meloidogyne se pusieron sobre un portaobjetos, y con una navaja de afeitar se cortaron en dos partes su cuerpo para eliminar su cutícula hasta formar un cuadro que contiene el patrón perineal, estos se limpiaron con ácido láctico al 45% durante 20 segundos y glicerina pura, los patrones perineales de Meloidogyne se montaron en lactofenol transparente. En el microscopio compuesto se observaron 20 patrones perineales para su identificación de especie de Meloidogyne, se comparó con claves morfométricas especializadas de Meloidogyne. Las características morfológicas que se observaron en los 20 modelos perineales, corresponden a estrías redondeadas en su parte dorsal y en su parte ventral, líneas laterales que dividen la parte dorsal y ventral que es característica distintiva para esta especie que corresponde a M. javanica. Esta investigación puede considerarse como pionera en la identificación de la especie de Meloidogyne en begonia en México.

Abstract:
This investigation presents the identification specie of root- knot nematode Meloidogyne, by the perineal pattern. The root-knot were collected in the begonia plant cultivar Cocktail under greenhouse conditions. Staining nematodes in root tissues by the Goodey method, after under the stereoscopic microscope the Meloidogyne female were removed from root fragment. Continued the females were cut in half of their body with razor thin, thereby eliminating exposure of eggs. The perineal patters were cleaned with 45% lactic acid for 20 seconds, and a drop of pure glycerin, it was mounted on clear lactophenol. Perineal patters were cut to a square. Four perineal patters for sample in a glass microscope slide, with a total of 20 preparation, were observed in light microscope for your identification. The identification of specie Meloidogyne were compared with specialized Meloidogyne morphometric keys. The perineal model of nematode root-knot was identified as M. javanica. Its distinctive feature is the lateral lines o lateral ridges that divide the dorsal and ventral striae. The dorsal arch is low and rounded. This research can be considered as one of the pioneers in the identification of the Meloidogyne species by method perineal pattern in begonia in Mexico
