EVALUACIÓN DEL EFECTO DEL ESTRÉS POR ZINC EN EL PERFIL FENOTÍPICO Y FITOQUÍMICO DE AGAVE SALMIANA CULTIVADO EN SISTEMAS DE INMERSIÓN TEMPORAL

Una estrategia importante para la conservación de las plantas del género Agave (Asparagaceae) reside en la diversificación de sus usos y su industria. Trabajos previos han posicionado a A. salmiana como una fuente prometedora de compuestos bioactivos de interés, como saponinas y flavonoles. El presente estudio tuvo como objetivo evaluar el efecto del estrés por zinc en el perfil fenotípico y fitoquímico de A. salmiana cultivado en sistemas de inmersión temporal. Para ello, se indujo estrés en las plantas por medio de diferentes concentraciones de ZnSO4 en el medio de cultivo (0, 50, 100 y 200 mM) y distintos tiempos de inmersión (1 y 5 min) durante 21 días. Posteriormente se tomaron mediciones físicas de las plantas. Se estimó su contenido de clorofila por medio de análisis de imagen. Por último, se prepararon extractos de las plantas y se evaluó su actividad antioxidante por ORAC, así como su contenido de saponinas y flavonoides por HPLC-ELSD-DAD. El crecimiento y desarrollo de las plantas se vio fuertemente inhibido por las concentración de ZnSO4. Asimismo, el contenido de clorofila de las plantas se redujo casi a la mitad al verse expuesta al zinc. La actividad antioxidante mostró ser 1.1 veces superior en el tratamiento de mayor exposición a zinc (200 mM y 5 min) respecto al promedio. No se consiguió evidencia suficiente para demostrar un cambio significativo en el contenido de flavonoles entre tratamientos. Respecto a las saponinas, estas disminuyeron drásticamente su contenido en al menos 28.9 % en las plantas sometidas a estrés. Este estudio realizó anotaciones sobre el umbral de toxicidad del zinc en A. salmiana. A partir de esta referencia, se pueden buscar concentraciones menos agresivas para elicitación.

    An important strategy on conservation of Agave (Asparagaceae) plants resides on the diversification of its uses and its industry. Previous works have positioned A. salmiana as a promising source of bioactive compounds of interest, such as saponins and flavonols. This study aimed to evaluate the effect of zinc stress on the phenotypical and phytochemical profile of A. salmiana plants grown on temporary immersion systems. For that, stress was induced in plants through different ZnSO4 concentrations in culture media (0, 50, 100, 200 mM) and distinct immersion times (1 and 5 min) for 21 days. After that, physical measurements of plants were taken. Their chlorophyll content was estimated by image analysis. Lastly, extracts of the plants were prepared and analyzed for antioxidant capacity by ORAC, as well as for their saponin and flavonol content by HPLC-ELSD-DAD. Growth and development of plants seemed to be severely inhibited due to ZnSO4 concentration. Furthermore, chlorophyll content was reduced almost by half when plants were exposed to zinc. Antioxidant activity showed to be 1.1 times higher in the treatment with the most exposure (200 mM and 5 min) than the average. There was not enough evidence to demonstrate any significant change in flavonol content among treatments. Regarding saponins, their content diminished drastically by at least 28.9 % in stressed plants. This study made annotations about the toxicity threshold of zinc in A. salmiana. With this reference, now it is possible to look for less aggressive concentrations for elicitation.
