EVALUACIÓN CITOGENÉTICA DEL EXTRACTO ACUOSO DE Taxodium mucrona-tum EN Vicia faba MEDIANTE MICRONÚCLEOS.

Estudios recientes confirman propiedades antimicrobianas y antitumorales de especies del género Taxodium, comúnmente conocido como “ahuehuete”, tule o “ciprés de Moctezuma”. En este contexto, el presente estudio aporta datos citogenotóxicos para futuras investigaciones farmacológicas. Como sistema de prueba, se utilizó el ensayo de micronúcleos en células meristemáticas de Vicia faba L., que analiza la frecuencia de micronúcleos (MN) y el índice mitótico (IM). Se utilizaron cuatro concentraciones de extracto acuoso de hojas: 10, 20, 50 y 100 μl/ml. Como testigo negativo se empleó agua destilada, mientras que el dicromato de potasio a 0.5 mg/ml fue el testigo positivo. Se realizaron cinco ensayos de repetición. Las raíces de V. faba expuestas en los tratamientos y en los testigos fueron cosechadas a las 24, 48 y 72 horas. Inmediatamente, se aplicó la técnica citogenética mediante tinción con acetoorceína para su observación al microscopio de células en división (%IM) y para el conteo de micronúcleos (%MN). Los datos promedio fueron analizados mediante la prueba paramétrica ANOVA (α < 0.05) y, al detectarse diferencias significativas, se sometieron a la prueba múltiple de Dunnett (α < 0.05). Previamente, se realizaron pruebas colorimétricas para identificar metabolitos secundarios en las hojas del árbol, identificándose los siguientes tipos de compuestos: saponinas, fenoles, flavonoides, glucósidos y taninos. El extracto acuoso de T. mucronatum no mostró diferencias significativas en el %IM respecto de la media del testigo negativo a las concentraciones de 10, 20 y 50 µl/ml. La concentración de 100 µl/ml fue citotóxica. Ninguna de las concentraciones mostró diferencias significativas en la frecuencia del %MN. En conclusión, el extracto acuoso de T. mucronatum presentó citotoxicidad leve sobre la división celular de V. faba y no provocó daño genotóxico.

  • Abstract:

    Recent studies confirm the antimicrobial and antitumor properties of species from the genus Taxodium, commonly known as “ahuehuete”, tule or “Montezuma cypress.” In this context, the present study provides cytogenotoxic data for future pharmacological research. The micronucleus assay was used in meristematic cells of Vicia faba L. to analyze the frequency of micronuclei (MN) and the mitotic index (MI). Four concentrations of aqueous leaf extract were used: 10, 20, 50, and 100 μl/ml. Distilled water was used as the negative control, while potassium dichromate at 0.5 mg/ml served as the positive control. Five replicate tests were carried out, and the roots of V. faba exposed to the treatments and controls were harvested at 24, 48, and 72 hours. The cytogenetic technique was then applied using acetoorcein staining to observe dividing cells (%MI) and count micronuclei (%MN) under the microscope. The average data were analyzed using parametric ANOVA (α < 0.05) and, when significant differences were detected, subjected to Dunnett’s multiple test (α < 0.05). Colorimetric tests were also carried out to identify secondary metabolites in the tree leaves, including saponins, phenols, flavonoids, glycosides, and tannins. The aqueous extract of T. mucronatum did not show significant differences in the %MI compared to the negative control at concentrations of 10, 20, and 50 µl/ml. The 100 µl/ml concentration was cytotoxic. None of the concentrations showed significant differences in the %MN frequency. In conclusion, the aqueous extract of T. mucronatum showed mild cytotoxicity on V. faba cell division and did not cause genotoxic damage.
